Loukyam opens with Venky (Gopichand) kidnapping the sister of Babji (Sampath Raj), as she is getting married to Bharat (Bharath Reddy) although she is in love with someone else. He goes to Hyderabad and waits for everything in his home, Warangal to clear. He stays with his friend, Shyamala (Satyam Rajesh). There he falls for Chandrakala, the sister of Sathya (Rahul Dev), a don in Hyderabad. Chandrakala rags other students in the college using her brother's power. After her initial rejection, Venky gets her to reciprocate his feelings. Meanwhile, Babji comes to Hyderabad in order to reach Sippy (Brahmanandam), the driver who helped Venky kidnap Babji's sister. Babji, unaware of the fact that he is travelling in Sippy's taxi, goes around Hyderabad with Boiling star Bablu (Prudhviraj), A rising Television star. Meanwhile, Kesava Reddy (Mukesh Rishi), a politician is behind Chandrakala's life. He conducts an attack on her through which Venky comes to know that Chandrakala is actually Babji's second sister and that he had sent her with Sathya to Hyderabad to maintain a low profile. Babji readies Chandrakala to marry Bharath. He also continues to search for whoever (Venky) abducted his sister. Using the help of Nano Shastri (Krishna Bhagavaan), Venky manages to get Babji and Chandrakala into his house. He also comes to know that the reason Kesava Reddy wants to kill Chandrakala is because his younger sister committed suicide because she loved Babji although he did not reciprocate the same. He hires Hamsa Sippy (Hamsa Nandini), Sippy's wife, to act as his father, Meka Papa Rao a.k.a Puppy's wife. He tells Babji's elder sister to come back on the engagement day to act as if not married and Babji fixes the marriage of his elder sister with Bharat. But unexpectedly, They all come to know that the girl is pregnant and so Bharat refuses to marry. Then Venky tells Babji that his brother (who is actually Babji's sister's husband and is acting as Venky's brother) will marry his sister. Babji agrees for the wedding and in turn tells Chandrakala that she is to marry Bharat again. Through an attack incident, Venky comes to know that Bharat is Kesava Reddy's son. He tells Kesava Reddy that even Babji is aware of this fact and is planning to kill Bharat on a trip to Srisailam. Kesava Reddy ends up believing him and attacks Babji and the group on the way. He foolishly reveals to Babji that Bharat is his son and that he is aware Babji will kill him. When Kesava Reddy realizes his folly, he kidnaps Chandrakala and Bharat and escapes from the spot. Boiling star Bablu who mistakes it to be a serial shoot stops Kesava Reddy on the way which gives Venky enough time to reach Kesava Reddy. In a thrilling climax, Venky gets Babji to beat up Kesava Reddy and using his "Sympathy angle" trick, which he had earlier used to get Babji and his sister together, He gets Babji to agree for his and Chandrakala's wedding as well. The film ends with Sippy saying that with "Loukyam"(Tact), you can fool not only Babji but even God and Boiling star Bablu saying he is Cool Bablu from now on and he will do only comedy serials and not action ones.

Kalyan Krishna (Shaam) comes to meet his old friend, Kalyan (Ravi Teja), who is now married to Naina (Ileana), settled in Virginia has become very rich and is still looking for a kick in life. When Krishna asks Kalyan about his son, Kalyan gets angry and says his son is worse than him. Robin Hood (Ravi Teja), is Kalyan's son and despite his father saying, Robin looks for "comfort" more than Kick. Robin was taken out of his mother's womb in only seven months because he felt it was uncomfortable. Robin became a doctor so that if he gets injured, he can treat himself immediately. Robin wishes to start a hospital in Hyderabad with their land, but Kalyan says that it has been grabbed by a goon, Settlement Durga (Ashish Vidyarthi). Robin arrives in India and stays in the rented house of Pandit Ravi Teja (Brahmanandam), but Pandit is continuously tortured by Robin. Robin also challenges Durga that he will get back his land without taking out a hand on him.

The film focusses on a family living in Hyderabad whose head is Ram Murthy (Nagineedu), a retired head master. He is a disciplined person and always tries to make his relatives behave in the same way. To avoid bad remarks from the society about his family, he sets up a family constitution of discipline. His mania for discipline is such big that if any one of his family members commit 100 Mistakes, that person would be banished from the family and that mistake can be anything which Ram Murthy hates which ironically includes laughing out loudly. Thus all of the family members maintain a disciplinary life to avoid being banished from the house. In detail, the family consists of Ram Murthy's wife Seetha, an Asthma patient; his daughter Kasthuri, his son in law Anand and their kids. Apart from them are Ram Murthy's two sons Brahmaji (Brahmaji) and Sundeep (Sundeep Kishan). Since Ram Murthy's brother Subrahmanyam (Prithviraj) was alleged to be committed 100 Mistakes though he didn't, he is sent away from home. While all of the family members commit mistakes less than 20, Sundeep commits 99 Mistakes and one more, he is out of the family. So to avoid that Sundeep starts leading a careful life. Brahmaji's marriage is scheduled at Tirupathi and the whole family boards Venkatadri Express at Kacheguda. Sundeep's mother forgot the Mangala sutra at the house and Sundeep goes away to bring it back. Moreover to his convenience, Sundeep is in S4 while others are in S3. So Sundeep goes outside the station and waits for an Auto-Rickshaw, But none accepts to go to his area. Meanwhile a miser techie Prarthana (Rakul Preet Singh) reaches Kachiguda railway station to board the Venkatadri Express and the Auto-Rickshaw driver goes to bring the change in order to pay her off the balance amount after receiving the fare. Sundeep gets into the vehicle and goes to his home. He breaks the house's lock with a stone as he has no keys to open it. He takes the Mangala sutra and goes back to Kacheguda Railway Station. Prarthana catches yet another auto and reaches to the station. There she pays off the fares of both the vehicles and goes to board the train.

Kalyan Sinha is a corrupt IPS officer who gets himself transferred to Hyderabad and misuses his power to gain monetary benefits in unorthodox methods. During this process, he also encourages a local MP named GK who has become a huge headache to Murali Krishna, the DGP of Hyderabad. Kalyan happens to be Murali's son who is angry on the latter as his supposed negligence killed his mother and just born sister, though Murali actually left his wife in the hospital to save several families in a riot. Unaware of this, Kalyan left Murali and joined an orphanage and became an IPS officer to seek revenge on his father. Subrahmanyam (Allari Naresh) is an intelligent, hardworking software engineer. He lives with his uncle (Ahuti Prasad), keeps the poster of Bill Gates in his room, spends most of his time with his girlfriend Madhu (Anita Hassanandani), and Busy Balraj (Brahmanandam). Krishna Murthy (Rajendra Prasad) is son of industrialist Satyanarayana (Nutan Prasad). Satyanarayana is worried about Krishna Murthy's marriage, so much so that he imagines every young woman who happened to be seen by him as his daughter-in-law. Krishna Murthy meets Padma (Rajani) in his friend's marriage and cupid strikes them. Krishna Murthy is afraid of revealing his love for Padma to his father directly as he fears his father may reject it out rightly. His initial attempts to reveal his love life to his father through letters are thwarted by his butler in fear for Satyanarayana's life. Desperate to reveal the love matter to Satyanarayana, Krishnamurthy then tries various gimmicks and at last is successful. A panicked Satyanarayana inquires about the family background of Padma and finds out that she is the daughter of Lakshmipathy (Kota Srinivasa Rao), a noted miser in his village. Aware of his father's concerns, Krishnamurthy reassures Satyanarayana and lands in Lakshmipathy's house posing as a miser. Krishnamurthy's pretense and pranks beget intended effect in Lakshmipathy and brings him closer to Padma. A marriage proposal from Sudhakar (Subhaleka Sudhakar), who is accompanied by his two glutton brothers much to the chagrin of Lakshmipathy, for Padma ends in Lakshmipathy favoring Krishnamurthy. Elated with the turn of events, Krishnamurthy prepares for marriage with Padma and invites Satyanarayana. However, his plans turn soar as Lakshmipathy, in his greed for money, abuses Krishnamurthy assuming that he is from poor family, before Satyanarayana confronts Lakshmipathy. A dejected Krishnamurthy requests Satyanarayana to give him one more chance to teach Lakshmipathy a lesson. Krishnamurthy, along with 'aragundu' Brahmanandam (Brahmanandam), a disgruntled butler of Lakshmipathy set about to transform Lakshmipathy. Krishnamurthy tacitly arranges for three different marriage proposals, starting with Sudhakar, with each proposer agreeing to offer more bride price than the earlier proposer. Overcome with greed for money, Lakshmipathy agrees to get her daughter married with each one of them. At the time of marriage with the bridegroom who offered the highest bride price, the other two bridegrooms also appear at the same time and question Lakshmipathy. An embarrassed Lakshmipathy is censured by Padma for his greed.
