The story is based on the concept of karma and bad deeds. Viren Sahai (Amitabh Bachchan), is an underworld don who rules over India. His son Abhir (Sushant Singh) is spoilt and believes he can do anything due to his father's power. When Viren's enemy Khan (Kader Khan) attacks Abhir, Viren demands revenge. Viren receives full proof information that Khan will be hidden at an local cinema theater on an particular date. The plot then moves onto the life of a simple cook, Shekhar Bhatia (Akshay Kumar) who is married to Dr. Kavita (Bhumika Chawla). Shekhar lives with his parents, wife and his younger brother Aryan (Aryeman Ramsay). When Aryan runs away from home, Shekhar searches the whole city to find him. During his search, he witnesses Viren attacking the cinema theater in which Khan is hidden, and decides to help the innocent people in the cinema.

The plot revolves around two fathers and their sons. Raghuvaran, Vikram's father, is a smuggler who lives in a house near S.P.Balasubramaniam's, father of Ajith. In his childhood days, getting attracted by Raghuvaran's activities, Ajith remains to stay with him always. While SPB tries to convince his son, it is all in vain. He got vexed and started to raise Vikram, son of Raghuvaran, as a good man. In college days, Vikram becomes a singer, and becomes a lover boy in the college. While Ajith studying in the same college becomes a dancer and turns out to be a local dada, who helps Raghuvaran in his business. In the meanwhile, Ajith and Vikram love the same girl (Maheswari), who in turn loves Ajith. SPB comes to know about Vikram's love affair and begs Ajith to leave his love for Vikram, since he won't be able to lead a peaceful life in the shape he is. The film ends with Ajith having to choose between his lady love and the dark world.

The movie begins with Amar (John Abraham) narrating the story of his family and himself. Amar is the son of Vidyadhar (Amitabh Bachchan) and Sumitra Patwardhan (Sharmila Tagore), a middle class couple in India. Amar lives and works in London and he sends some of his salary to them. One day Amar returns home with Jenny Mayer (Anusha Dandekar), his girlfriend, and announces his intention to get married to her. Initially reluctant, his parents are pleased and happy for the two. The film is set in British India in the year 1915. The story is about a young doctor, Govardhan Menon (Mohanlal), who is wrongly accused of bombing a train carrying 55 people including British officials. He is put in a cellular jail at Port Blair, Andaman and Nicobar Islands. Kaalapani shows the sufferings of hundreds of Indian prisoners in the cellular jail, including leading participants of the freedom movement. The extremely inhumane conditions faced by the prisoners in the jail are accurately depicted.
