Elle commence sa carrière en 1995 dans Barsaat pour lequel elle reçoit le Filmfare Award du meilleur espoir féminin. Elle s'illustre également à Tollywood. En 2001, après son mariage avec l'acteur Akshay Kumar, elle décide de mettre fin à sa carrière.
Le 14 janvier 2001, elle épouse l'acteur Akshay Kumar, puis la même année donne naissance à son premier enfant, Aarav. Le 12 juillet 2012, Rajesh Khanna décède.

Twinkle Khanna est la fille du célèbre acteur Rajesh Khanna et de l'actrice Dimple Kapadia, respectivement d'origine punjabi et gujarati. Elle est également la sœur de Rinke Khanna et la nièce de Simple Kapadia, toutes deux actrices. Twinkle Khanna étudie dans un pensionnat à Panchgani, puis elle intègre l'université Narsee Monjee de Mumbai.Le 14 janvier 2001, elle épouse l'acteur Akshay Kumar, puis la même année donne naissance à son premier enfant, Aarav. Le 12 juillet 2012, Rajesh Khanna décède.

Inspector Suryadev Singh (Amrish Puri) is an honest and efficient cop in the city. He has a granddaughter Kajal (Twinkle Khanna), whom he has brought up after her parents were poisoned by his enemies, and whom he values more than his life. But Suryadev’s cousin Vishambar (Suresh Oberoi) is plotting the complete ruin of Suryadev, along with his wife and his brother in law Bhanwari (Shakti Kapoor) because Vishambar’s deceased father gives all his share of property, including a village Sundernagar, to Suryadev as Vishambar has married someone against his wishes. So Vishambar plans to kill Kajal, as then his revenge will be complete, and the property will fall into his hands since there would be no one to inherit it from Suryadev, which his son Nagendra can inherit. For this purpose, Vishambar hires Karan (Ajay Devgan).

Nisha lives a very wealthy lifestyle in Neelgaon, India, with her businessman dad, Sardar Roop Singh. She is now of marriageable age, and her dad wants her to marry his friend's son, Sohan, but Nisha dislikes him. While traveling to Darjeeling with a dance troupe, she meets with her dad's business associates' son, Popat Lal, and after a few misadventures, both fall in love with each other. She takes him to meet her dad where he can also finalize his business transaction, but when Roop comes inside, he finds that Popat has disappeared, and in his place is another man claiming to be the real Popat. Nisha's heart is broken and she starts to hate Popat. She does meet with Popat, who tells her that his real name is Sunder and both had been promised to each other by their respective parents, but Roop had subsequently changed his mind. Nisha believes him and agrees to marry him without her father's blessings. When the marriage is to take place, a man named Khanna comes over and tells Nisha that Sunder is already married to woman named Shanti, who he subsequently killed, and had been the primary accused in this case by the police. The story follows Mansukh (Himesh Reshammiya), the son of a marriage bureau owner Champak Lal (Manoj Joshi (actor)). Since he has grown up, Mansukh tries to help his father in the business, however all the alliances he had tried to get done have resulted in separation even before the marriage took place. To prove his worth to his father, he takes up an unusual challenge of getting an underworld don Tatya Tukaram Tendulkar's (Mithun Chakraborty), spoilt brat sister Indu (Asin Thottumkal) getting married to a cop called Bahattar Singh (Akshay Kumar) aka Khiladi 786 in Punjab. He convinces Tatya to pretend to be a cop to make Bahattar agree to the marriage. But little did the over enthusiast Mansukh know that Bahattar Singh, along with his father Sattar Singh (Raj Babbar) and his uncle Ikhattar Singh (Mukesh Rishi) were not cops but a family of conmen. The story takes a shape as the two hardcore families pretend to be policemen in front of each other and the beans from each side can be spilled anytime which can be life threatening for Mansukh. And to make the situation worse, it turns out that Indu has a boyfriend, Azad (Rahul Singh), who is a criminal locked up in jail. When Bahattar finds out, he helps Azad escape from jail to make Indu happy. But no sooner does Indu realize that she truly loves Bahattar.

The film revolves around a struggling road constructor, Sachin Tichkule (Akshay Kumar). He dreams big, but there seems to be no chance of his dreams coming true as he doesn't have any money to bribe the bureaucracy. His family have lost faith in him, telling him to earn money. To make matters worse, the new Municipal Commissioner turns out to be his former girlfriend, Gehna Ganpule (Trisha Krishnan), who now hates him. His brothers-in-law Trigun (Manoj Joshi), Suhas Vichare (Milind Gunaji), and his elder brother (Paritosh Sand) were all responsible for a bridge collapsing case in which many people were killed. They were helped by a politician named Sanjay Rana (Jaideep Ahlawat). After being saved by their driver Vishwas Rao, (Tinu Anand) they kill him fearing he would reveal the truth. Meanwhile Sanjay's lustful eyes fall on Sachin's sister Anjali (Urvashi Sharma). Sachin warns him by slapping him in his office. Meanwhile Azad Bhagat (Makrand Deshpande) wants justice as his family were killed in that accident. Anjali is married to Sanjay, unbeknownst to Sachin, when he confronts his father he reprimands him, saying he has no right to say he didn't know anything because he has no money to send his sister off. After losing contact, he learns she has been attempting to contact him and goes to their house, seeing Azad escape from Sanjay's house. He then learns of Anjali's death because a cooker exploded. Devastating the whole family, especially Sachin himself, who is suspicious. He then meets Azad again at the municipal office, he learns of Azad's intentions and that he had stolen evidence which would put the people involved, behind bars. They both then agree to help each other to reveal the corruptness behind the construction of the bridge. Sanjay hires masked robbers, who kill Azad, who was on his way to court. At the hospital, while Azad is dying in Sachin's arms, he reveals that while he was stealing the evidence, he saw Sachin's sister being raped by Sanjay and his friends, now speculating whether she was murdered or if she committed suicide. In an attempt to make sure the evidence isn't revealed, Sanjay steals it after robbing Azad's house, a fight ensues between Sanjay and Sachin, Sanjay is then killed after being run over and Sachin successfully retrieves the evidence. Sachin's brother and brothers-in-law are arrested, and the whole family begin belittling him, claiming he was jealous of them and unlike his successful brothers, and blaming him for tarnishing the family name. Sachin then argues back that, they didn't lose anyone and he had to deal with the pain of losing Anjali the most. He agrees to leave, until his father stops him and realises his honesty and trustworthiness, re-kindling their relationship.
