Irandam Ulagam is about two stories which unfold concurrently in two different planets – our Earth and an unnamed exoplanet. On earth a timid medical student Ramya (Anushka Shetty) who has a crush on an academic mathematician Madhu Balakrishnan (Arya). He initially refuses her proposal for marriage as his father is in valid needing his full attention and care. Later, Madhu gradually reciprocate Ramya's love seeing her caring nature and proposes marriage. But, she reluctantly rejects his proposal as her family had already arranged marriage. While on a trip to Goa, Madhu wins Ramya's heart while her fiance abruptly cancels the marriage citing dowry as reason. She instantaneously dies in a freak accident and this is followed by the news of death of Madhu's paralysed father too. Depressed by the sudden twin tragedies, Madhu becomes a vagabond in Goa. He sees an apparition of his dead father that tells him death is an illusion. Later, in one of the tourist spots, he follows an empty car to a hilltop and tries to commit suicide. He gets injured and faints while trying to drive the car.

Karthik Swaminathan (Dhanush), referred to as 'Genius' by his friends, is a freelance photographer who aspires to become a wildlife photographer like Madhesh Krishnasamy (Raviprakash). His close friends Sundar (Sunder Ramu), Shankar (Mathivanan Rajendran), Padmini (Pooja Devariya) and Vindhya (Shilpi Kiran) support him and his sister after the death of their parents. One day, Sundar introduces Yamini (Richa Gangopadhyay) as his girlfriend to Karthik and his friends. On their first meeting, Karthik and Yamini have a bitter experience but they slowly develop a special bond. Karthik tries to stay away from the girl since he does not want to break his friend's heart, but a sequence of events brings them closer. Eventually, Sunder's dad arranges for Karthik and Yamini to be married.

In 1279 A.D., the downfall of the Chola dynasty seems imminent as the Pandyas drive the Chola people out of their kingdom in southern India. To escape them and save the life of his successor the Chola emperor sends his son along with the Chola people to a secret territory. The refugees take along an idol sacred to the Pandyas, angering them. To capture the escaped Cholas and the stolen idol, the Pandyas extend their invasion to unexplored territories but cannot find them.

At the Kanyakumari beach, Geetha (Shriya Saran) looks at a lonely lying personal diary where she enjoys a poem written in it. She admires the poem by replying on the same page. While she and her friend admire the beauty of Sunrise, she accidentally loses one of her anklets (it falls in the sea). She sees a person jumping in after it and she perceives him to have died in the waters. The incident haunts her in her dream again and again and feels very guilty for it. In her college, she meets a rich, handsome boy Arjun (Sameer Dattani) who impresses her to win her love. He proposes her and threatens her to love him or else he would die by jumping from college roof top. Already feeling guilty for being cause of a person's death, not willing to let it happen again Geetha accepts his love and screams 'I love You' in front of the entire college to Arjun which was witnessed by Kutty (Dhanush).

Shakthivel (Bharath) is the son of a wealthy industrialist and the managing director of Rajasekar Industries. Disgusted with life, he gets up and walks aimlessly, taking a bus to the Chennai Railway Station. There he boards the Pandian Express at night without a ticket and sits in gloom. Anjali (Tamannaah) is a college student who is going to her hometown Theni for vacation. Anjali is bubbly and talkative and thoroughly irritates Shakthi. She tells him of her plans to elope with her lover Gautham. At night, Shakthi leaves the train. Anjali wakes up to find him gone. Seeing him sitting on the platform, she gets off the train to bring him back. As a result she misses the train. She tells Shakthi that it is his responsibility to take her back to the train. Annoyed, he takes her to the next station so that she can board the train and leaves her. Anjali again misses the train. While she is waiting for a taxi to reach the next station, some gangsters chase her, but she runs into Shakthi who fights them off.

The first few scenes themselves set the tone; first, a hesitant and bashful Venugopal (Cheran) trying desperately to carry on an interesting conversation with a prospective bride Ranjitha (Vimala Raman), in her picturesque home in Nagercoil. The conversation proceeds quite well in the beginning, until it hits an unexpected snag—Venu was hospitalised for extreme stress during his school years; he still stammers as the result of the strain, and when he's unexpectedly put into stressful situations.
