Singing cowboy Gene Autry (Gene Autry) is out of work and looking for a job. After pawning his guitar, he meets former rodeo sharpshooter Angie Burke (Vera Marshe), who works for wealthy ranch owner Estrellita Estrada (Elena Verdugo). Angie suggests that Gene ride across the border to El Sombrero Grande, Estrellita's ranch. Gene learns that the ranch is managed by James Garland (Stephen Dunne), an acquaintance from their days with Monahan's Wild West Show. Gene heads south into Mexico.

Échappé de la prison de Santa Fe, le bandit Wolf Benson monte dans un train en cours de marche. As president of the Flagpole Cattlemen's Association, singing cowboy Gene Autry (Gene Autry) entrusts the sale of the association's large cattle herd to young Jimmy Agnew (Rand Brooks), who is so thrilled with the opportunity that he proposes marriage to his girlfriend, Millie Jackson (Linda Leighton). The next day, while traveling to Hays City with the cattle, Jimmy is persuaded to join a poker game by two crooked gamblers, Dixie Trambeau (Tristram Coffin) and Joe Crowley (John Berkes), who fleece the naive cattleman and force him at gun point to turn over the power of attorney authorizing them to sell the cattle. Ashamed at his blunder, Jimmy goes into hiding.

After rounding up a large herd of horses in Texas, Gene Autry (Gene Autry) receives a telegram from his brother Tex (Ken Cooper), who is mining diamonds in South Africa. The message states that Tex and his partner, Edward Barclay, have discovered a big strike in the Valley of Suspicion and urges Gene to bring as many horses as possible to Dunbar, South Africa, where he can contact John Cardigan (LeRoy Mason), the saloon owner who grubstaked Tex and Barclay. Gene and his sidekick Frog (Smiley Burnette) quickly arrange for passage overseas and accompany the horses to South Africa.

A rash of strange cattle rustlings have occurred in which cattle are slaughtered on the range and their carcasses taken away. Sheriff Matt Doniphon (William Farnum) and his deputies, Gene Autry (Gene Autry) and Frog Millhouse (Smiley Burnette), watch over one rancher's cattle as they are driven to Box Canyon. After the sheriff and his men leave, the rustlers move in, radio the cattle's location from an airplane, and then bring in refrigerator trucks. The rancher and one of his workers are murdered, the cattle killed, and the carcasses taken away.
